Trans Problems

I dont know if anyone had this problem I cant find anything in forum about it, I had changed the tires to the Ofna Pirate tires and changed the pinion gear to 16 tooth to unload the motor, the trans shifted fine!!! Now I put the stock tires on the car and with the same 16 tooth pinion and the motor screems and the trans dont shift. I tried adjusting the trans shift point and still dont get a shift from the trans. Is the trans shifting on wheel revolution or motor rpm..... Thanx for any help anyone can give me, and what is the factory setting on the trans shift screw
